Es la concentración del antibiótico requerida para impedir el crecimiento bacteriano a partir de la incubación de 10 5-6 bacterias en fase de crecimiento rápido, en un medio libre de proteínas con pH 7,2, aerobio, durante un periodo de incubación de una noche. Este término es importante porque se utiliza para determinar la sensibilidad bacteriana a un agente antibiótico específico. Es importante recordar que las condiciones in vivo son distintas a las utilizadas para esta prueba que se realiza in vitro. En un ser vivo la bacteria generalmente se encuentra un medio más ácido y anaerobio. Además es mayor tamaño del inoculo bacteriano y probablemente no está en fase rápida de crecimiento; lo cual disminuye el valor predictivo del MIC.
